With its unparalleled resolving power for simultaneously visualizing hundreds of proteins in complex biological samples, 2-DGE has been used extensively to characterize wheat endosperm proteins 25 . Historically, 2-DGE methodology was first developed to separate wheat gliadins in order to study genotypic variation 26, 27 . In the original method, IEF was used for the first dimension and acid PAGE (lactate) for the second dimension 26 . Addition of carrier ampholytes, urea, and non-ionic detergent to the solubilization buffer and the first dimension gel together with SDS and reducing agent to the second dimension gel greatly improved the resolution of 2D gels 28-30 . 2-DGE continues to be utilized to analyze gluten proteins and assign chromosomal location of genes 31-33 . Cultivar Identification

Wheat grain proteomics is being investigated as an efficient method for identification of cultivars. In a preliminary study, Skylas et al. 44 found significant differences in the protein patterns in the HMW-GS, gliadin, and smHSP regions of 2D gel maps for four cultivars differing in flour quality. These authors concluded that a proteomics approach, especially when applied to the analysis of non-gluten proteins, opens up new possibilities for the development of immunoassays for cultivar identification. In a study comparing two closely related commercial varieties that differed in processing characteristics, Skylas et al. 45 identified 23 proteins that could be used to distinguish the cultivars. Yahata et al. 46 compared the protein maps of seven wheat cultivars and identified 25 proteins, 21 of which were gliadins and glutenins, that could be used as cultivar-specific markers.

Antiinflammatory Diets

An alteration or loss of regulation of the AA cascade leads to a chronic inflammatory state, which characterizes numerous physical disorders. A frequently indicated offender to be removed from the diet is conventionally raised red meat, a significant source of AA. It should be clarified that grass-fed beef has a higher omega-3 to omega-6 ratio than conventionally raised beef. Additionally, white meat from poultry and eggs may be high in AA depending on the type of feed. Another source of dietary inflammation is hydrogenated foods, which are high in the pro-inflammatory trans-fatty acids. Numerous studies highlight a link between foods that are high in omega-6 fatty acid and decreased intake of omega-3 fatty-acid-rich foods.6 Dietary gluten and lectins are also recognized as common triggers of inflammation.

Treatment

Limited evidence suggests that dietary restriction of gluten and casein may be beneficial in children with autism. A small controlled study found that such children showed significantly better development with a gluten- and casein-restricted diet than did controls.54 Food allergy may play a role in autistic symptomatology, as suggested by significantly increased levels of antibodies to casein, beta-lactoglobulin, and lactalbumin in autistic children than in controls, followed by reductions in behavioral symptoms with an eight-week elimination diet for these three food components.55 However, a small, double-blind, placebo-controlled study failed to find any significant changes in autistic children with a gluten- and casein-restricted diet, although the children's parents did report improvements in the children's conditions.56 Another study found behavioral improvement after six months in 60 of autistic children given a ketogenic diet.57

Wheat and Barley

Although detailed studies of the distribution of gluten proteins in wheat have not been reported, evidence currently available indicates that similar differences in distribution of S-rich, S-poor and HMW components may exist to those reported in barley. In particular, the use of the GUS reporter gene to study promoter activity in the developing grain of transgenic lines shows clear differences in the patterns of expression conferred by promoters from LMW subunit (St ger et al. 2001) and HMW subunit (Lamacchia et al. 2001) genes. This clearly shows that the LMW subunit promoter is most strongly expressed in the sub-aleurone cells and outer part of the starchy endosperm and the HMW subunit promoter in the central part of the starchy endosperm these distributions are consistent with the analyses of barley discussed above.
