Hydrate 3 g of dried ground maize seeds in a mortar in 10 mL of water for 30 minutes, at room temperature. Then, add 20 mL of acetone and leave for 15 minutes. Extract carotenoids by grinding the mixture in a mortar and pestle with 50 mL acetone. Vacuum filter the residue in a Buchner funnel equipped with filter paper Whatman # 2. Return the residue to the mortar and repeat the procedure until the residue is nearly colourless, usually once more. Transfer one-third of the filtrate to a separating funnel containing 20 mL of petroleum ether, to which 300 mL of distilled water is added. Discard the aqueous layer. Repeat the procedure for the remaining filtrate. Wash the organic phase 3 times with 200 mL of distilled water and pass through 15 g of anhydrous sodium sulfate into a round bottom flask. Concentrate the sample with a rotary evaporator and dry (Rodriguez-Amaya and Kimura, 2004).
